Its something I have been interested in but need to learn more about. I would probably need 2 maps. One for premium fuel and one for E85. Who here uses it?

I use it in my GT500, it's equivelant to 105 octane and burns very cool. Only downside to it though is its Stoich value drops from 14.64:1 (E0) / 14.06:1 (E10) to 9.76:1 (E85) for cruising and for your WOT fueling for a FI car it would need to go to about a 7.5:1 Air:Fuel, so the lambda would still be the same.

Nice advantage to it is that you can pretty much run a NA timing profile on a FI car. You'll need to make sure your tank is pretty damn empty though if you switch over to your E85 map and make sure you bring a tester with you when you go for gas, because some gas stations like to cheapen out and mix it, and you end up with E65 and E42.5... which can cause issues.

I gained over 95rwhp/90tq on my Shelby from switching 93 to E85

Anything good enough for unleaded gas is good enough for E85. What you do need to know if you have a fuel system capable of supporting E85 as it does pump about 30% more fuel over a standard unleaded gas system.

So if youre planning to make XXX HP and your fuel system is close to meeting it's max, plan for 30% higher.
